Pilot scale production of hypercin and hyperforin in hairy root cultures of Hypericum perforatum in bioreactor

Hypericum L. species are distributed throughout several countries in the world. Among the numerous species, Hypericum perforatum L. has been used in traditional medicine for over two thousand years and nowadays is mainly used for the treatment of depression and neurological disorders. Hypercin and hyperforin are two main secondary metabolites of this specie. For medicinal purposes, plants require approximately 2 years to concentrate the metabolites in the flowers and unfortunately the cultivars with enhanced secondary product accumulation are susceptible diseases. Total hypercin and hyperforin are challenging in the field-grown plants depends on environmental conditions. The aim of this project is development of an optimized culture media and condition for hairy root culture of Hypericum perforatum to produce hypercin and hyperforin in pilot scale using bioreactor. The project consists of three sub project. The transcriptom and genome of this plant subjected to be study under different treatments to recognize and differentiate the key enzymes and transcription factors involve in hypercin and hyperforin biosynthesis. We also would like to manipulate for metabolite pathway to achieve an enhanced level of hypercin and hyperforin in hairy roots. Design of appropriate bioreactor for mass production of hairy roots is another research interest in this project.
